Since updating software to 5.0 on Galaxy S5 - my Juice Defender app will not work and battery just drains away within hours. Any solutions please???

Try a factory reset - don't forget to back up your content and make sure your contacts and calendar are fully sync'd first. Samsung devices are particularly sensitive to bits of the old code hanging round memory and confusing the system and a reset will get rid of them.

Google have made a lot of changes to the way the system works, some of which would actually break apps like this. Project Volta and the ART are now standard in 5.0 both of which should massivly improve battery life and performance on any device running Android 5.0. This in mind there shouldn't really be a need to use apps like Juice Defender anymore.
Was there a particular feature it provided that you are missing? There may be an alternative that could be used that won't harm the system.

On the basis of what Nabs has said, I'd uninstall Juice Defender first and see if that improves the situation. You may save yourself from the rigours of a reset.
