My S6 won't charge at all using the lead...but will using the wireless charger. So this tells me it's something to do with the actual charging port that the lead...

Take it to your nearest Samsung Repair Centre and get them to check out the device for you. Based on what you are saying, it sounds like a fault with your charging port. They will be able to advise if this is the case and will be able to repair the device. I've used them myself and found them to be pretty good and helpful. They were able to sort out the issue with my S4 in just a few hours!
I would imagine so - unless the reason it isn't working is down to physical damage. The only reason I raise that point is that physical damage is always excluded from warranty claims.
